If you save the Combi you can load it into the insert intact. If it's an instrument, you will need to patch the inputs of the Combi into something - anything (like a Spider that's not patched to anything). I've always heard that a common method is 1) EQ (cut) > 2) Compression > 3) EQ (boost/shape/cut) with #1 being optional and used to prevent unwanted frequencies from triggering the compressor and being amplified by it.
